The lyophilization process consists of three key stages: freezing, primary drying, and secondary drying. The nucleation of ice crystals, the initial step in ice formation during freezing, traditionally unfolds spontaneously and over an extended period. However, our innovative LYOSPARK® technology has changed the game by allowing for controlled nucleation at specific temperatures and rates, enhancing batch homogeneity and reducing cycle duration.

In the past, the random nature of nucleation made lyophilization a challenging process. For example, when filling pharmaceutical formulations in vials, crystals could form at temperatures ranging from -5 to -20 °C. The nucleation temperature directly influences the ice structure, crystal size, and the appearance of the dried product, impacting Critical Quality Attributes (CQAs) such as residual moisture and dissolution behavior.

2 LYOSPARK® — Controlled Nucleation.

LYOSPARK® — SIMULTANEOUS CRYSTAL NUCLEATION.

Integrating LYOSPARK® technology into conventional freeze-drying processes is straightforward. Vials are loaded into the freeze dryer and cooled to the optimal nucleation/ cooling temperature. Once the nucleation temperature is reached, the chamber is vented with gas from the freeze dryer’s condenser.

A filter-equipped cooling trap is attached to the chamber, where ice crystals form on the cooled surface and are carried away in the gas stream during venting. Inside the supercooled product solution, these ice crystals trigger an immediate and homogeneous nucleation in each vial. The nucleation and the subsequent formation of ice crystals at higher temperatures results in larger pores, reducing drying times. LYOSPARK® offers superior scalability and higher throughput speeds, optimizing performance and productivity.

Process Optimization LYOSPARK® eliminates issues such as active ingredient denaturation and unpredictable scale-up. It offers numerous technical advantages, including enhanced product quality, consistent vial uniformity per batch, and, depending on the product, a more uniform, aesthetically pleasing cake.
